If you want to get the right sound you have to know how to tune a guitar. You can do it by hand, which a lot of people do, the only problem is that if the low E string (6th string) is not tuned correctly, you could end up overtightening and snapping it.
I prefer using electric guitar tuners. You can get them for $20-$30 for a pretty solid one. Zzounds.com has some pretty good ones, like the Fender AX12 Chromatic Tuner, which is a pretty good one for about $30.
